Description

The TGF-beta family members comprise of about 40 signaling proteins that play important regulatory roles in embryonic development and adult tissue homeostasis. These signaling molecules are bone morphogenetic proteins (BMPs), growth and differentiation factors (GDFs), anti-Mullerian hormone (AMH), activin and nodal. Bone morphogenetic protein 11 (BMP11), also known as growth and differentiation factor 11 (GDF11) is a TGF-beta signaling component and is involved in many physiological process including bone formation, neurogenesis, embryonic skeletal patterning, post natal bone formation and metabolism, cell cycle and cell arrest etc. BMP11/GDF11 is highly expressed in mesodermal tissue during embryogenesis (1)s and regulate the axial skeletal patterning as well as limb skeletal formation (2). BMP11 protein is initially translated as precursor protein that undergoes proteolytic cleavage to generate the mature BMP11 protein and BMP11 propeptide corresponding to C and N-terminal peptides respectively (3). The BMP11 propeptide has been shown to antagonize theBMP11 activity by forming latent complex with BMP11 (2). Such latent interaction with TGF beta signaling proteins expands the range of TGF-beta like proteins to form such complexes and widen their physiological functions.

The Myostatin (Mstn) and BMP11/GDF11 are highly related transforming growth factor B family members that are involved in regulating embryonic development and adult tissue homeostasis. Despite being closely related and have high degree of sequence homology, targeted mutations in these genes resulted in non-overlapping phenotype affecting distinct biological process. The mutations in Mstn in mice resulted in doubling of skeletal muscle mass while loss of BMP11/GDF11 caused a dramatic anterior transformations of the axial skeleton, kidney agenesis, and increase in progenitor cell number (4).

BMP11/GDF11-selective antibodies were generated against a peptide taken from the human BMP11/GDF11 protein spanning amino acids from 60-100. The BMP11/GDF11 -selective antibodies are affinity purified on an immobilized antigen based affinity matrix, the isolated antibodies were then stabilized in antibody stabilization buffer for long-term storage.
